invasion Immunisation of such molecules can create antibodies that will block the entry of the parasite in the host cells Several such molecules are being tested for their vaccine potential Most of these molecules differ in structure depending on plasmodium species and strains and their protective effects are limited Share This Article Related Article While working on a protein called Enolase we discovered that its primary function is to burn glucose in living organisms to produce energy We also found it to be present on the surface of the parasites that invade red blood cells Since Enolase is normally found inside a cell we wondered what was it doing on the surface and tried to see what happens if we block it with anti-Enolase antibodies We made this protein in the laboratory and immunised mice with it and found that it protected the animals against malaria We observed that the antibodies isolated from the immunised mice when added to parasite culture led to inhibition of parasite growth As this protein is also present in humans and has similar structure antibodies generated against the whole molecule could be detrimental to host The challenge was to identify the part of parasite which is required for invading red blood cells In a recent publication we identified a small segment in the amino acid sequence of the protein which is unique to plasmodium This segment about five amino acid long is not found in human beings We added this small fragment to nano-particles and used them for immunisation of mice Results showed it can protect against malaria As this unique segment of amino acids is present in all species of plasmodium so a vaccine developed along these lines would be equally effective on all species Secondly the parasite cannot change the molecular structure of the Enolase protein because it is crucial for glucose burning and release of energy Recent findings from other laboratories showed the presence of Enolase on the surface of ‘ookinete’ a stage of the parasite that invades mosquito gut Antibodies against Enolase inhibit this invasion process also It raises the possibility that newly identified vaccine candidate sub-structure may also block transmission through mosquitoes Our research has shown an alternative way of attempting to develop a vaccine for malaria Of course there is a long way to go But our findings have the potential to provide a vaccine that could be effective against multiple strains and species of parasite that cause malaria in humans and also block transmission through mosquitoes
